GC101 Phase III Trial Meets Primary Endpoint in Spinal Muscular Atrophy

2026-10-29

On October 15, 2026, Genecradle Therapeutics announced that its investigational AAV-based gene therapy, GC101, met both the primary and key secondary endpoints in a confirmatory phase III trial for 5q spinal muscular atrophy (5q SMA).

GC101 is the first AAV-based gene therapy developed in China for intrathecal treatment of 5q SMA. Administered as a single intrathecal injection, it delivers a gene encoding functional survival motor neuron 1 (SMN1) directly to the central nervous system to enable sustained SMN protein expression and improve motor neuron function by addressing the underlying cause of the disease. GC101 has previously received Breakthrough Therapy Designation from China’s Center for Drug Evaluation and Orphan Drug Designation from the US FDA.

Currently, two AAV-based gene therapies for SMA-Zolgensma and Itvisma-are commercially available. According to the report, this trial is the first SMA gene therapy study worldwide to utilize a randomized, head-to-head superiority design against an existing treatment. This milestone represents a decisive step toward advancing GC101 from clinical development to potential commercialization and underscores continued progress in AAV-based gene therapies for genetic neuromuscular disorders.

As part of the broader gene therapy ecosystem, Genevoyager provides end-to-end CRO/CDMO services, from drug discovery to large-scale manufacturing, leveraging both HEK293 and our proprietary Bac/Sf9 AAV production platforms to produce high-quality AAV vectors with minimal impurities and no detectable rcAAV. With 30+ drug development projects and multiple IIT/IND submissions supported, Genevoyager has helped advance AAV-based gene therapies and recombinant protein vaccines into clinical development, including programs targeting Parkinson’s disease, refractory epilepsy, hepatitis B, and cancer, with selected programs receiving FDA IND clearance and entering Phase I trials. Backed by extensive experience, we are committed to delivering safe, high-quality solutions to academic, pharmaceutical, and biotech partners worldwide.
